چکیده

Hyaluronan-binding function of the CD44 molecule has not been so far detected in myeloid cells. To study pure populations of primitive myeloid cells, we investigated the hyaluronan-binding function of the CD44 molecule from three myeloid cell lines: KGla, KGI, and HL60. Both KGla and KG1 cells express the CD34 antigen characteristic of the hematopoietic stem cells and HL60 cells do not; accordingly, KGla and KG1 cells are generally considered as the most primitive and HL60 cells as the most mature of these cell lines. Measurement of cell adhesion to hyaluronancoated surfaces (using 6‘Cr-labeled cells) and of aggregate formation in hyaluronan-containing solutions, showed that 45% of KG1 cells and 22% to 24% of KG1 a spontaneously bind to hyaluronan, whereas HL60 cells do not either spontaneously or after treatment with a phorbol ester. Hyaluronan binding by KGla and KG1 cells is mediated by CD44, because it is specifically abolished by monoclonal antibod-

P-130: Designing and Construction of An Appropriate Eukaryotic Expression Vector to Generate Soluble Form of Human Hyaluronidase Type PH20 in Cell Culture Feasible for Application in IVF and ICSI

Background: The hyaluronidases are the enzymes hydrolyze β-1, 4 glycosidic linkage of hyaluronan. Hyaluronan is a polymer consisting of a repeating disaccharide unit found in cumulus ovuforus complex, semen liquid and other tissue. Addition to hydrolyzing the hyaluronan, hyaluronidase can penetrate through the cumulus cells layer that surrounds the oocyte, thus it terms spreading factor. Moreov...

O-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells

Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...

Evaluation the Efficacy of HBA Sperm- Hyaluronan Binding Assay Method in Infertility Centers

Introduction: The novel development of the (HBA) sperm- hyaluronan binding assay has now been routinely used in some laboratories worldwide to predict sperm maturity and functionality. The purpose of this study was to evaluate the correlation between HBA score and histone deficiency, protamine content and DNA fragmentation. Methods: During April- March 2012 patients who were enrolled at ...

Binding of function-blocking mAbs to mouse and human P-selectin glycoprotein ligand-1 peptides with and without tyrosine sulfation.

P-selectin glycoprotein ligand-1 (PSGL-1) mediates rolling of leukocytes on P-selectin-expressing endothelial cells under shear flow. Function-blocking monoclonal antibodies (mAbs) against mouse and human PSGL-1 recognize an anionic segment at the N-terminus of PSGL-1. High affinity interaction of PSGL-1 with P-selectin requires sulfation of tyrosines 46, 48, and 51 (human) or 54 and 56 (mouse)...
